"She's going to kill it," Melissa McCarthy says of the first-time Emmy host

Jane Lynch already has practice winning an Emmy. But come Sunday night, she’s going to face a new challenge: running the show.

Fellow costars and nominees say the Glee star and first-time Emmy host, who is also nominated for her role as Sue Sylvester, is definitely up for the gig.

“She’s going to kill it,” Bridesmaids star Melissa McCarthy, who is an Emmy nominee for her role on Mike & Molly, tells PEOPLE. “And I have a good seat to watch Jane just destroy it.”

Dot-Marie Jones, who costars with Lynch on Glee, says she’s been clocking plenty of practice time as she gears up for her hosting gig.

“She’s been rehearsing forever,” Jones tells PEOPLE. “I saw her one day at work for, like, five minutes. I’m so excited for her.”

And costar Iqbal Theba, who plays Principal Figgins on the Fox show, says Lynch is nothing like her prickly character – not that he doesn’t want to see a glimmer of Sue’s attitude on Sunday’s telecast (which airs on Fox, 8 p.m. ET, live from the Nokia Theatre).

“She will be the best host. She puts her heart and soul in everything she does,” Theba says. “I hope there’s a splash of Sue there.”

As for her chances at collecting a second Emmy?

“I think she’ll win,” he says. “I’m going to cry when she wins.”
